A second American Ninja Warrior has been announced by G4 for SASUKE 26. Tryouts were held in Windward Plaza at Venice Beach, California on August 7 and 8. Also at the events were SASUKE All-Star Nagano Makoto, Shin Sedai Urushihara Yuuji, and Okuyama Yoshiyuki. Each were there to sign autographs and all three got to test out the obstacles and try out the qualifying course for themselves. Both Okuyama and Urushihara passed, but Nagano failed the jump to the Jumping Bars.

The competition was broken up into three sections: the Venice Beach course which ran the same as American Ninja Warrior's Qualifying and Semifinal rounds, the California Mountains which was a five-day boot camp, and the Japan course (the same as SASUKE 26 but G4 edited their broadcast to showcase only their qualifiers). The competition was taped in August 2010 and aired on three consecutive Wednesdays in December 2010. The Japan runs aired on December 23, 2010, ten days before the official airing of SASUKE.

Venice Beach

As with its predecessor, two rounds of the Venice Beach course were run. The competition spanned over three days. The first two days consisted of the Qualifier round where the top thirty times were taken from the first six obstacles of the course. The third day consisted of the Semifinal round where the aforementioned thirty challengers ran the full nine obstacle course. The top fifteen results (in terms of progress using time from the previous obstacle as a tiebreaker) moved on to the California mountains course.

California Mountains

The top fifteen had to spend five days at a reality-style boot camp which contained replicas of the Warped Wall, Floating Boards (only shown during training time; never attempted in a relay), Cliffhanger, Balance Tank, Double Salmon Ladder, Circle Slider and Slider Jump to determine the final ten who will attempt SASUKE 26. The fifteen were split into three teams of five; the Red Dragons (Furlanic, Campbell, Hall, Steffensen and Kasemir), the White Tigers (Meeuwenberg, Daniels, Cousins, King, and Laplante) and the Blue Monkeys (Orosco, Cusic, Cedars, Dollard, and Truesdell). The teams went through two challenges daily: the Pressure Challenge and Team Relay. The Pressure Challenge consisted of each team nominating a member to outlast the others in whatever challenge was for that day. The worst performed team would then be subject to a "punishment" which makes them lose training time for the race. In the relay, each member does two of the selected obstacles for that day while carrying extra weight when running to the obstacles; failing completely would result in a time penalty for the overall team time. The team with the highest overall time would then nominate two of their members (in the case the team cannot come to a decision, the winning team will have to) to race on the the "Heavenly Ropes", a replica of the Tsuna Nobori/G-Rope from the Final Stage. The first person to the top or the person who made it furthest (as was the case with Patrick Cusic vs. Dorian Cedars) would be safe, and the loser was eliminated.

Japan Course

As per tradition, the top ten competed on SASUKE 26 with a $250,000 reward for anyone who is able to achieve kanzenseiha. Prior to SASUKE 26, Levi Meeuwenberg severely injured his wrist and had to forfeit his position in the competition. Levi would pass on his chance to California Mountains Boot Camp finalist Adam Laplante. Despite this setback, Levi attended SASUKE 26 as a sideline spectator and morale boost for the American competitors.
